目的探讨血小板活化因子受体(PAF-R)在重症急性胰腺炎(SAP)胰腺组织的动态变化及银杏苦内酯B(BN52021)干预前后的影响。方法Wistar大鼠随机分为阴性对照组(NC组)、SAP模型组(SAP组)、BN52021治疗组(BN组),每组按手术后不同时相点(1、2、3、6、12和24h)分为6小组,分别测定血淀粉酶的变化,并对胰腺组织进行病理学观察和评分,用Western blot检测胰腺组织PAF-R蛋白质表达的变化。结果BN52021能降低SAP的血淀粉酶和改善其病理变化:SAP组和BN组胰腺组织PAF-R在3h达到高峰(P〈0.05),除1h外其余各时相点与NC组有显著性差异(P〈0.05):BN组与SAP组之间无显著性差异。结论PAF-R在SAP过程中显著升高.对SAP的病情发展有重要意义,BN52021对PAF-R蛋白表达无显著影响,可能通过竞争性抑制上升的PAF与表达量同时上升的PAF-R相结合,实现它的治疗作用。

英文摘要:

Objective To explore the dynamic change of platelet- activating factor receptor(PAF-R) in pancreatic tissue of rats with severe acute pancreatitis(SAP) and the effect of ginkgolide(BN52021) on in- tervention of SAP. Methods Wistar rats were randomly divided into three groups, negative control group( NC group), SAP model group( SAP group) and BN52021 treating group( BN group). The rats of each group were further randomly divided into six subgroups according to different postoperative time points ( 1, 2, 3, 6, 12, 24h). The activity of blood plasma amylase was determined with entire automatic biochemistry appearance. Meanwhile, observe the pancreatic tissue in pathology and grade it according to pathology standardization. The change of quantity of PAF-R protein in pancreatic tissue was determined by Western blot. Results BN52021 can degrade hemodiastase and improve the pathological change in SAP. It was a kind of dynamic change in the detection of PAF-R protein in pancreatic tissue between SAP group and BN group. The quantity of PAF-R pro- tein reached peak at 3h time point(P 〈0.05). There was a significant difference in the detection of PAF-R in pancreatic tissue between SAP group and NC group or BN group and NC group at different time points(P 〈 0. 05) except at lh. There was no significant difference between SAP group and BN group at different time points (P 〉 0.05). Conclusion SAP model group may be successful. The quantity of PAF-R protein is increased significantly in the process of SAP, which will promote development of SAP. BN52021 can not affect the quan- tity of PAF-R protein significantly. It may ameliorate rats SAP via competitive inhibiting the binding of rising PAF and rising PAF-R of SAP.
