一氧化氮供体硝普钠对猪腔前卵泡体外培养的影响

研究在培养液中添加不同浓度(0.000、0.001、0.010、0.100、1.000 mmol/L)一氧化氮供体硝普钠(sodium nitroprusside,SNP)对猪腔前卵泡体外培养的影响。结果表明,SNP对猪腔前卵泡存活、发育和腔的形成、卵母细胞正常发育有一定促进作用,但高浓度会产生抑制作用。体外培养后,各处理组卵泡直径差异不显著(P〉0.05);处理第4天,0.001 mmol/L SNP处理组卵泡存活率(85.28%)显著高于1.000 mmol/L SNP处理组(70.60%,P〈0.05);0.001 mmol/L SNP处理组卵泡成腔率(79.81%)和卵母细胞正常率(50.93%)显著高于1.000 mmol/L SNP处理组(55.22%,35.00%,P〈0.05);0.001 mmol/L SNP处理组COC回收率(23.27%)著高于其余处理组(P〈0.05),其余各处理组差异不显著(P〉0.05);1.000 mmol/L SNP处理组颗粒细胞凋亡率显著高于其他各组(P〈0.05),而0.001 mmol/L SNP处理组颗粒细胞凋亡率显著低于其他各组(P〈0.05),其余各组间差异不显著(P〉0.05)。

英文摘要:

To understand the effects of sodium nitroprusside(SNP),an NO donor,on the growth and development of porcine preantral follicles in vitro culture,follicles were treated with different concentrations of SNP(0.000,0.001,0.010,0.100,1.000 mmol/L).Results showed there were no significant variances(P0.05) in the follicular diameter between all treatments during in vitro culture;the survival rate in 0.001 mmol/L SNP treatment was significantly higher than that in 1.000 mmol/L SNP treatment(85.28%vs70.60%,P0.05);the antrum formation and normal rate of oocyte reached highest in 0.001 mmol/L SNP treatment(79.81%,50.93%),significantly higher than that in 1.000 mmol/L SNP treatment(79.81vs55.22,50.93%vs35.00%,P0.05);the recovery rate of cumulus cells oocyte complexes(COC) in 0.001 mmol/L SNP treatment was significantly higher than that of control and any other treatments(23.27 vs 15.00 vs 13.89 vs 11.67 vs 8.33,P0.05),there were no significant difference between other treatments(P0.05);the granulose apoptosis in 1.000 mmol/L SNP treatment was significantly higher than that in other treatments(P0.05),while that in 0.001 mmol/L SNP treatment was significantly lower than that in other treatments(P0.05),there were no significant differences between other treatments(P0.05).Certain SNP was beneficial to the survival,development,antrum formation of preantral follicle and normal development of oocytes,while high level of SNP has toxic effect.
