裸卵的体外培养对于动物繁殖和人类辅助生殖具有重要的意义。实验研究成熟培养基中维生素C和维生素E的添加对猪裸卵(denuded oocytes,DOs)体外发育能力的影响。将维生素C以0、50、100、250、500μmol/L和750μmol/L的浓度,维生素E以0、10、20、50、100μmol/L和250μmol/L的浓度加入成熟液中,比较猪DOs的体外成熟率、孤雌激活后的卵裂率和囊胚发育率。结果表明:维生素C促进猪DOs的体外成熟和囊胚发育;维生素E对猪DOs的成熟率无影响,增加了囊胚率。综上所述,维生素C和维生素E可提高猪DOs的体外发育能力。
The in vitro culture on cumulus cells free oocytes is of the great importance theoretically and practically to animal reproduction and human assistant reproduction. The experiment investigated the effect of vitamin C and vitamin E on in vitro developmental potential of porcine cumulus cells denuded oocytcs (DOs). In two independent experiments, porcine DOs were cultured alone supplemented with different concentrations of vitamin C (0, 50, 100, 250, 500 μmol/L and 750 μmol/L) or vitamin E (0, 10, 20, 50, 100 μmol/L and 250 μmol/L), respectively. The maturation rates, cleavage rates and blastula rates after parthenogenetic activation were compared respectively. It was demonstrated that vitamin C promoted the in vitro maturation and the development of blastula of porcine DOs; vitamin E didn't increase the in vitro maturation rates, but improved the blastula rates. In summary, both vitamin C and vitamin E can enhance the in vitro developmental potential of porcine DOs.
