目的:探讨Peroxiredoxin Ⅲ(PrxⅢ)在大鼠超负荷心肌肥厚过程中的表达变化。方法:雄性SD大鼠随机分为假手术对照组和模型组,模型组大鼠实施腹主动脉缩窄术,制备压力超负荷心肌肥厚模型。分别于术后24h,2、4、6周,测定血液动力学变化;称量心体质量;光镜和电子显微镜观察左心室的组织变化;提取左心室RNA,RT-PCR分析PrxⅢ的表达变化。结果:术后24h两组大鼠的血液动力学和心肌形态无明显变化。术后2、4、6周时,模型组大鼠的SBP、DBP、LVSP、LVED及左心室体质量指数逐渐增加,心肌纤维逐渐增粗,线粒体逐渐增多,肌节增长。术后24h PrxⅢ的表达稍有降低,但无统计学意义,2周时则明显降低,4周有所上升但仍低于对照,6周时明显上升且高于对照组。结论:PrxⅢ mRNA表达呈现先降低后增高的趋势,即在心肌肥厚中早期表达降低,而在心肌肥厚形成后表达明显升高,表明PrxⅢ参与了超负荷心肌肥厚的发生。
Objective: To investigate the expression change of peroxiredoxin Ⅲ(PrxⅢ) in process of rats overload cardiac hypertrophy. Methods: Male SD rats were divided randomly into a shame operation control group and a model group. Cardiac hypertrophy model was made through partial ligation of the abdominal aorta. After 24 h, 2 weeks, 4 weeks, 6 weeks of operation, physiological index was first detected, then left ventricle was observed under light and electron microscopes and PrxⅢ mRNA expression was measured. Results: Model of rat cardiac hypertrophy was formed on 6 weeks after operation through analyzing the change of the physiological index and morphology. Compared to the control group, the expression of PrxⅢ mRNA was lowered on 24 h and further lowered on 2 weeks. PrxⅢ mRNA began to enhance but was still lower than that of control on 4 weeks; On 6 weeks, PrxⅢ mRNA was significantly higher than that of the control group. Conclusion: The expression of PrxⅢ first decreased and then increased during the process of rat pressure-overload cardiac hypertrophy. PrxⅢ may take part in the forming and development of pressure-overload cardiac hypertrophy.
