目的:研究氧化苦参碱在食蟹猴体内的药物动力学及代谢转化率。方法:建立食蟹猴血浆中氧化苦参碱及苦参碱的液相-串联质谱(LC/MS/MS)测定法;测定ig和iv氧化苦参碱后氧化苦参碱和代谢产物苦参碱的血药浓度,以及ig和iv苦参碱后苦参碱的血药浓度;计算氧化苦参碱的药动学参数与代谢转化率。结果:氧化苦参碱的代谢转化率为(30.7±6.6)%。结论:氧化苦参碱在食蟹猴体内代谢转化率高。
OBJECTIVE To study the pharmacokinetics of oxymatrine (OMT) in cynomolgus monkeys and its metabolic conversion rate. METHODS The LC/MS/MS method was established to detect OMT and its active metabolitc matrine(MT) in plasma obtained from cynomolgus monkeys; some plasma concentration- time curves were used to calculate the pharmacoki netic parameters of OMT and its metabolic conversion rate after the administration of oMT or MT through iv and ig. RESUL'IN OMT metabolic conversion rate in cynomolgus monkeys was (30. 7 ± 6. 6)%. CONCLUSION OMT metabolic conversion rate in cynomolgus monkeys was high.
