目的观察食管鳞癌组织中Wnt2、散乱蛋白(Dvl)mRNA和蛋白的表达变化,并探讨其意义。方法取60例食管鳞癌患者肿瘤组织标本为观察组,癌旁组织标本为对照组。用RT-PCR法检测两组Wnt2、Dvl mRNA相对表达量;用Western blot法检测两组Wnt2和Dvl蛋白表达量。结果观察组Wnt2 mRNA、Dvl mRNA、Wnt2蛋白、Dvl蛋白表达量分别为1.007±0.237、1.008±0.220、0.512±0.406、1.218±1.082,对照组分别为0.877±0.119、0.891±0.196、0.153±0.189、0.505±0.358,观察组Wnt2 mRNA、Dvl mRNA、Wnt2蛋白、Dvl蛋白表达量均高于对照组,P均〈0.05。观察组Wnt2与Dvl mRNA表达呈正相关(r=0.807,P〈0.01)。结论食管鳞癌组织中存在Wnt2及其通路关键蛋白Dvl mRNA及蛋白的高表达。Wnt信号通路在食管鳞癌的发生、发展中发挥了重要作用。
Objective To observe the expression changes of Wnt2 and dishevelled( Dvl) mRNA and protein in the esophageal squamous-cell carcinoma( ESCC) tissues,and to investigate the significance. Methods Sixty cases of tumor tissue specimens( observation group) and adjacent tissues( control group) from ESCC patients were selected. The mRNA and protein expression of Wnt2 and Dvl in the two groups was respectively detected by RT-PCR and Western blotting. Results In the observation group,the mRNA expression of Wnt2 and Dvl was respectively 1. 007 ± 0. 237 and 1. 008 ±0. 220,the protein expression of Wnt2 and Dvl was respectively 0. 512 ± 0. 406 and 1. 218 ± 1. 082,and that in the control group was respectively 0. 877 ± 0. 119,0. 891 ± 0. 196,0. 153 ± 0. 189 and 0. 505 ± 0. 358. The mRNA and protein expression of Wnt2 and Dvl in the observation group was higher than that of the control group( all P 0. 05). In the observation group,Wnt2 and Dvl mRNA was positively correlated with each other( r = 0. 807,P 0. 01). Conclusion The high expression of Wnt2 and Dvl gene is found in ESCC tissues,which indicates Wnt signaling pathway plays an important role in the occurrence and development of ESCC.
