目的探讨散乱蛋白(Dvl)基因在食管鳞癌发生、发展中的作用。方法采用RT-PCR和Western blotting法检测60份食管鳞癌组织及其相应癌旁正常组织中Dvl mRNA及其蛋白的相对表达量,分析其与食管鳞癌患者临床病理特征的关系。结果食管鳞癌组织及其癌旁组织Dvl mRNA的相对表达量分别为1.008±0.220、0.891±0.196(P<0.01),Dvl蛋白相对表达量分别为0.478±0.152、0.261±0.079(P<0.01)。Dvl mRNA相对表达量与食管鳞癌患者淋巴结转移、TNM分期有关(P均<0.01),Dvl蛋白相对表达量与食管鳞癌患者浸润深度、淋巴结转移、TNM分期有关(P均<0.01)。结论 Dvl基因可促进食管鳞癌的发生、发展,其高表达提示TNM分期较晚、易发生淋巴结转移。
Objective To investigate the function of mRNA and protein expression of Dishevelled ( Dvl) gene in the occurrence and development of esophageal squamous carcinoma ( ESCC) .Methods We detected the expression levels of Dvl mRNA and protein in 60 cases of ESCC tissues and its corresponding adjacent tissues by RT-PCR and Western blotting , and then the relationships between the expression and clinical pathological features were analyzed .Results The expression of Dvl mRNA in the ESCC tissues and adjacent tissues was 1.008 ±0.220 and 0.891 ±0.196 (P<0.01), and the ex-pression of Dvl protein was 0.478 ±0.152 and 0.261 ±0.079, respectively (P<0.01).The Dvl mRNA expression was related with lymph node metastasis and TNM stages (all P<0.01), the expression of Dvl protein was associated with the depth of infiltration, lymph node metastasis and TNM stages (all P<0.01).Conclusion The Dvl expression promotes the occurrence and development of ESCC , its high expression indicates the late TNM stage and it is prone to the occurrence of lymph node metastasis .
