目的观察将在体记录扩散性抑制的电生理学方法应用于局灶性脑缺血后远隔缺血区(海马)的电生理学改变。方法采用局部给予3mol/L高浓度氯化钾溶液,光化学血栓形成局灶性脑缺血模型和大脑中动脉栓塞局灶性脑缺血模型诱导扩散性抑制;抑制组预先30min腹腔注射MK-801(0.2mg/100g);将记录电极加以改良置入海马记录细胞外直流电位变化。结果诱导组记录到幅度约10—30mV,时程大于1—2min的扩散性抑制波,抑制组未记录到SD波。并观察到大脑中动脉栓塞脑缺血模型诱导的海马扩散性抑制波。结论SD波可能参与MCAO模型中海马远隔效应的发生。
Objective By applying the electrophysiological recording method of spreading depression (SD) in vivo to study the electrophysiological changes in the region remote from ischemia area of focal cerebral ischemia. Methods SD waves were elicited by 3 mol/L high concentration potassium chloride solution, photochemical thrombotic focal cerebral ischemia and middle cerebral artery occlusion (MCAO) cerebral ischemia. In the inhibited group ,MK-801 (0.2 mg every 100 g body weight) was intraperitoneally pre-injected. The Ag/AgCl recording electrode was modified and applied to electrophysiological study in hippocampus of MCAO model. Results SD waves with an amplitude of about 10-30 mV and a duration of more than 1 rain were recorded in the induced group and no SD wave was detected in the inhibited group. SD was proved to exist in hippocampus after MCAO ischemia. Conclusion SD may play a part in remote effects of hippocampus in MCAO rats.
