2型糖尿病动物模型KK-Ay小鼠糖尿病肾病的鉴定

目的研究2型糖尿病KK-Ay小鼠糖尿病肾病(DN)的特点和应用价值。方法选择雄性KK-Ay小鼠和C57BIM6J小鼠各30只,每两周代谢笼收集24h尿液及随意尿测尿微量白蛋白,眶后静脉丛采血测空腹血胰岛素、糖化血红蛋白、血糖、胆固醇、甘油三酯、尿素氮、肌酐、白蛋白。8、12、20周龄时两组分另4处死10只小鼠取肾脏行病理检查。结果不同周龄KK-Ay小鼠尿微量白蛋白、血胰岛素、糖化血红蛋白、血糖、胆固醇、甘油三酯均比同周龄C57BIM6J小鼠高(P〈0.05),且随周龄增加而进展,8周龄表现为典型2型糖尿病,12周龄进入早期DN,20周龄肾脏病理表现为特征性DN改变,但未见肾功能下降。结论KK-Ay小鼠是研究DN早期病变的理想模型。

英文摘要:

Objective To investigate the application value and the feature of diabetic nephropathy in KK-Ay mice with type 2 diabetes. Methods Thirty male KK-Ay mice and thirty C57BL/6J mice were studied, 24-hour urine and urine at any time were collected by metabolism cage every two weeks, blood was collected from vena orbitalis posterior, uria trace albumn and insuline were detected by ELISA, whole blood glyeosylated hemoglobin was tested by capillary eleetrophoresis. Serum glucose, eholesterin , triglyceride, urea nitrogen, ereatinine, albumn were measured by automatic biochemistry analyzer. Ten mice of each group were sacrificed at the age of 8, 12, 20 weeks. Renal tissue was taken, and the renal pathology was tested. Results The urine trace protein., insulin, glyeosylated hemoglobin, serum glucose, eholesterin, triglyeeride in KK-Ay mice of each age of week were higher than those of C57BL/6J mice at the same age( P 〈 0.05 ) , and which were increased with the adding age of week, the KK-Ay mice developed into typical type 2 diabetes after the age of 8 weeks, and suffered early stage of diabetic nephropathy at the age of 12 weeks. At 20-week-age,renal pathology showed the typical dia- betic nephropathy,but renal function did not decrease. Conclusions The KK-Ay mice were confirmed to be a good animal model for early stage of type 2 diabetic nephropathy.
