目的观察艾灸补髓能否促进老年学习记忆减退大鼠海马神经发生。方法选取督脉“百会”和“命门”两穴对学习记忆减退SD老年大鼠进行艾灸治疗,腹腔注射5溴-2脱氧尿苷(BrdU)后以免疫荧光双标记技术观察BrdU阳性和BrdU与神经丝巢蛋白(Nestin)双阳性的细胞,结合细胞增殖与NSCs特异性标志物表达评价海马神经发生。结果艾灸能增加老年学习记忆减退大鼠海马BrdU阳性细胞的数量,提高BrdU和Nestin双阳性细胞的表达。结论提示艾灸具有促进老年学习记忆减退大鼠海马神经发生的作用。
Objective To investigate whether mairow-supplementing therapy of moxibustion can enhance hippocampal neurogenesis in aged cognitive decline rats. Methods Moxibustion therapy were performed on "Baihui" ( DU20 ) and "Mingmen" ( DU4 ) acupoints in SD aged rats. 5-bromodeoxyuridine(BrdU) were intraperitoneal injected before rats were sacrificed. Both BrdU and nestin positive labeled neurons in hippocampus were observed under immunofluorescent microscope to evaluate hippocampal neurogenesis combining with cell proliferation and neural stem cell specific markers. Results Positive neurons either single labeled or double labeled by BrdU and Nestin were increased in moxibustion group. Conclusions Moxibustion can increase hippocampal neurogenesis in aged cognitive decline rats.
