目的探讨脑梗死患者经丹参注射液诱寻重组人粒细胞集落刺激因子(rhG—CSF)动员自体外周血干细胞后,外周血脑源性神经生长因子(BDNF)表达情况。方法将60例14d后40岁~60岁发病14d后脑梗死患者分为丹参注射液+rhG—CSF组、rhG—CSF组、丹参组、常规组,每组15例。常规组予常规治疗;丹参组:丹参注射液20mL/d静脉输注,连用7d,余治疗同常规组。rhG—CSF组:在抽血检查次日08:00皮下注射古粒芬300μg,连用3d,余治疗同常规组。丹参+rhG—CSF组:第1天开始连续使用丹参注射液20mL/d静脉输注7d,同时在丹参注射液滴注第2天08:00皮下注射rhG—CSF300μg,连用3d,余治疗同常规组。4组均以7d为1个疗程。Elisa法检测外周血各组BDNF含量。结果丹参+rhG—CSF组及rhG—CSF组治疗后外周血BDNF表达分别为8.944ng/mL±1.932ng/ITIL,8.684ng/mL±1.412ng/mL,呈上升趋势,与丹参注射液组及空白组(7.874ng/mL±1.934ng/mL,7.309ng/mL±1.779ng/mL)相比,差异有统计学意义(P〈0.05)。结论丹参注射液配伍rh—CSF以及rh—CSF本身能够提高脑梗死患者外周血BDNF的表达,可能对脑梗死患者神经功能修复有一定的促进作用。
Objective To explore the effect of Danshen injection on brain - derived neurotrophic factor (BDNF) expression paracrine - secreted by autologous peripheral blood stem cells mobilization using recombinant human granulocyte colony - stimulating factor (rhG- CSF) in patients with cerebral infarction. Methods Sixty patients with onset of 14 days after cerebral infarction were divided randomly into four groups:Control group, rhG - CSF group given rhG - CSF 300 μg/d subcutaneously for consecutive 3 d at the next day after blood tests,Danshen injection group given Danshen injection 20 mL/d intravenously for 7 d, Danshen injection plus rhG- CSF group. There was 7 d for a course. The expression of BDNF in peripheral blood was detected by ELISA. Results After treatment, the expression of BDNF in peripheral blood were increased in Danshen injection plus rhG- CSF group(8. 944 ng/mL± 1. 932 ng/mL) and in rhG - CSF group(8. 684 ng/mL± 1. 412 ng/mL), which were higher than that in Danshen injection group(7. 874 ng/mL±1. 934 ng/mL) and control group(7. 309 ng/mL±1. 779 ng/mL,P〈0.05) ,respectively. Conclusion Danshen injection plus rh-CSF or rh - CSF could upregulate the expression of BDNF in peripheral blood,which could promote recovery of nerve function in patients with cerebral infarction.
