安徽省某鸡场120日龄蛋鸡发生疑似鸡传染性喉气管炎(ILT),根据Gen Bank发表的鸡传染性喉气管炎病毒(ILTV)株g D基因设计合成1对特异性引物,从发病鸡喉头及其分泌物中扩增出1 134 bp大小片段。将病鸡喉头气管及其分泌物处理后接种SPF鸡胚,接种120 h后在绒毛尿囊膜(CAM)上形成典型痘斑,证实分离到1株鸡传染性喉气管炎病毒,命名为AH-XY15株。应用鸡胚传代的第4代病毒感染5日龄健康麻羽鸡,结果感染鸡出现了典型ILT临床症状,并能够从发病鸡喉头分泌物检出ILTV。应用PCR扩增分离株毒力基因TK基因和g B基因,序列分析显示,AH-XY15分离株TK、g B基因的核苷酸序列与国内WG株、国外USDA株以及国内疫苗株K317株同源性均在99%以上,氨基酸序列同源性在97%-99.8%之间。
A strain of infectious laryngotracheitis virus isolate was obtained from larynx material and upper respiratory tract secretions of infected chickens with clinical respiratory signals and reduction of egg production.The virus isolate was identified by PCR amplification of specific g D gene sequence and formation of opaque plaques on the chicken chorioallantoic membrane(CAM) after serial embryonated egg passages,then it designated as AH-XY15.The pathogenicity test showed that the isolate caused typical respiratory symptoms to infectious laryngotracheitis on 5-day-old chickens post-challenge by AH-XY15 strain.The TK gene and g B gene of the AH-XY15 isolate were sequenced and analyzed.The AH-XY15 isolate shares the nucleotide homology of 99 %and the amino acids homology of 97%-99.8%with the reference of domestic WG strain,domestic vaccine strain K317,and abroad USDA strain on TK gene and g B gene.
