目的对藏方甲嘎松汤挥发油指纹图谱的特征峰与体外抗氧化活性进行谱效相关性研究,为其药效评价和合理应用提供初步实验依据。方法通过气相色谱质谱联用技术建立甲嘎松汤挥发油的指纹图谱,采用DPPH和ABTS+自由基清除方法探讨甲嘎松汤挥发油体外抗氧化作用,通过皮尔逊相关分析、灰关联度分析对特征峰与药效进行谱效学分析。结果依据谱效相关性大小,确定单个抗氧化作用成分贡献的大小顺序依次为对聚伞花烯、(-)-4-松油醇、桉油精、α-雪松烯、α-姜烯、1R-α-蒎烯、α-松油醇、石竹烯、榄香素、肉豆蔻醚、β-水芹烯、α-蒎烯。结论该研究确定了甲嘎松汤挥发油指纹特征峰抗氧化药效的大小,为甲嘎松汤药用物质基础研究,药效评价及质量标准制定提供参考。
Objective To investigate the relationship of fingerprints of essential oil from Tibetan medicine Jiagasong Decoction with antioxidant activities,thus to provide a reference for the evaluation criteria of its pharmacodynamics and rational uses. Method The fingerprints of essential oil from Jiagasong Decoction were established by gas chromatography-mass spectrometry, and the antioxidant effects were evaluated according to the scavenging of2, 2-diphenyl-1-picryl-hydrazyl(DPPH) and 2, 2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) ABTS+radicals. The relationship of characteristic absorption band of fingerprints with the pharmacological action was calculated by Pearson relational analysis and grey relational analysis. Result According to the relational sequence of characteristic peaks, the essential components contributing to the antioxidant activities in decreasing order were as follows:p-cymene,terpineol-4,eucalyptol,α-cedrene,α-zingiberene,1R-α-pinene,α-terpineol,caryophyllene,elemicin, myristicin, β-phellandrene1, α-pinene. Conclusion The study has revealed the order of fingerprint characteristic peaks of Jiagasong Decoction essential components contributing to antioxidant activities, which will provide evidence for the pharmacological substance of Jiagasong Decoction,and for its pharmacodynamics and quality control.
