目的从保护肝脏线粒体和调控"Nrf2/Bach1-ARE-抗氧化酶"通路两个角度,观察三味干姜散对实验性肝损伤动物模型的影响。方法复制大鼠急性酒精性肝损伤模型和小鼠刀豆蛋白A(Con A)免疫性肝损伤模型,分别给予三味干姜散后,检测肝功能指标ALT、AST,进行病理学观察;提取肝线粒体,检测线粒体膜电位、MPTP、SOD、8-OHd G、呼吸链复合物Ⅰ、Ⅲ酶活性以及肝组织中MDA、GSH、GSH-Px水平;采用RT-PCR检测肝组织中Nrf2、Bach1、HO-1的m RNA表达水平。结果对于急性酒精性肝损伤模型,三味干姜散能降低ALT、AST(P〈0.05)和8-OHd G(P〈0.01)以及MDA的含量(P〈0.05,P〈0.01),提高肝组织中抗氧化剂GSH、SOD和GSH-Px酶的活性(P〈0.05,P〈0.01),上调Nrf2和HO-1 m RNA的表达(P〈0.05),显著增加呼吸链复合物Ⅰ、Ⅲ酶的活性(P〈0.01),以及降低线粒体膜通道孔的通透性(P〈0.05);对于免疫性肝损伤模型,三味干姜散能降低ALT、AST水平和MDA活性(P〈0.05,P〈0.01),升高SOD(P〈0.05)、GSH-Px与GSH酶的活性(P〈0.05,P〈0.01),显著上调线粒体膜电位(P〈0.01)。结论三味干姜散对大鼠急性酒精性肝损伤和小鼠Con A免疫性肝损伤均有一定的保护作用,其机制与三味干姜散抗肝脏线粒体损伤和调控"Nrf2/Bach1-ARE-抗氧化酶"通路有关。
Objective To evaluate the effect of Sanwei Ganjiang Powder on animal models of experimental liver injury through observing its protective effect on hepatic mitochondria and its regulation of Nrf2/Bach1-ARE-antioxidase pathway.Methods We established rat model of acute alcoholic liver injury and mice model of Con A-induced liver injury,and then the model rats and mice were given Sanwei Ganjiang Powder.After treatment,we detected indicators of liver function of alanine transaminase(ALT)and aspartate transaminase(AST),observed the hepatic pathology,extracted the liver mitochondria for the detection of mitochondrial membrane potential,1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP),superoxidase(SOD),8-hydroxy-2-deoxyguanosine(8-OHd G)and respiratory chain complexes Ⅰ,Ⅲ enzyme activity,and examined the hepatic malondialdehyde(MDA),GSH,and GSH-Px levels.Meanwhile,the hepatic m RNA expression levels of Nrf2,Bach1 and HO-1 were detected by RT-PCR.Results In the model of acute alcoholic liver injury,Sanwei Ganjiang Powder could decrease the levels of ALT,AST(P〈0.05) and8-OHd G(P〈0.01)and MDA(P〈0.05,P〈0.01),SOD and GSH-Px activities(P〈0.05,P〈0.01),up-regulated the expression of Nrf2 and HO-1 m RNA(P〈0.05),significantly increased the activities of respiratory chain complexⅠ and Ⅲ(P〈0.05,P〈0.01),and decreased mitochondria(P〈0.05).In the model of immune liver injury,Sanwei Ganjiang Powder can decrease the levels of ALT,AST and MDA(P〈0.05,P〈0.01),increase SOD(P〈0.05)GSH-Px and GSH enzyme activity(P〈0.05,P〈0.01),significantly increased mitochondrial membrane potential(P〈0.01).Conclusion Sanwei Ganjiang Powder has protective effect on rats with acute alcohol liver injury and mice with Con A-induced immunological liver injury,and its mechanism is probably related to the counteraction of hepatic mitochondrial damage and with the regulation of Nrf2/Bach1-ARE-antioxidase pathway.
