目的研究胃癌中转录因子Snail及黏附因子E—cadherin的表达,并探讨与胃癌浸润转移的关系。方法用组织芯片和免疫组化sP的方法检测189例胃癌组织、54例癌旁组织、32例正常胃黏膜中Snail及E—cadherin表达。结果胃癌组织Snail阳性率f84.13%)显著高于癌旁组织f48.15%,10〈0.05),Snail的表达与胃癌不同分化程度、浸润深度、淋巴结转移、TNM分期及Lauren分型有关fP〈0.05)。胃癌组织E—cadherin的阳性率(22.2%)显著低于癌旁组织(55.5%,P〈0.05),E—cadherin的表达与胃癌不同分化程度、浸润深度、TNM分期及Lauren分型有关(P〈0.05)。E—cadherin阳性组生存期高于阴性组,Snail阴性组生存期高于阳性组。结论Snail蛋白高表达与E—adherin蛋白低表达可能参与胃癌浸润转移的发生,并且联合检测可作为其重要生物学标志,有助于胃癌早期诊断和预后评价。
Objective To investigate the expression of Snail and E-cadherin in gastric cancer and evaluate their relationship between tumor invasion and metastases. Methods Immunohistochemical staining was adopted to detect the expressions of snail and E-cadherin on tissue microarray in 32 cases of gastric normal mucosa, 54 eases of Para-Cancerous Tissue, and 189 cases of gastric cancer. Results The rate of Snail positive cells in Gastric cancer was 84.13%, significantly higher than that of P mucosa, 48.15%, (P〈0.05). The expression of Snail is related with the differential degree, the depth of tumor invasion, lymphoid metastases and distant metastases (P〈0.05). The rate of E-eadherin positive cells in Gastric cancer (22.2%) was significantly lower than that of Para-caneerous mueosa (55.5%, P〈0.05). The expression of E-cadherin is related with the differential degree, the depth of tumor invasion, lymphoid metastases, TNM stage, and Lauren types (P〈0.05). The survival of E-eadherin positive group is higher than that of negative group, while the survival of snail negative group is higher than that of positive group. Conclusion E-eardherin-positive expression, but not Snail, may be associated with a better prognosis. The combined detection of Snail and E-eadherin contributes to early diagnosis and the evaluation of prognosis.
