目的观察基质细胞衍生因子-1(SDF-1)在脑缺血-再灌注损伤大鼠血清中的表达情况及通窍活血汤对其影响。方法雄性SD大鼠30只,按随机数字表法随机分为假手术组、模型对照组及通窍活血汤小、中、大剂量组各6只,建立大鼠脑缺血-再灌注损伤(MCAO)模型。造模成功后,通窍活血汤组给予通窍活血汤灌胃,其余组给予纯化水灌胃。应用酶联免疫吸附测定法(ELISA)检测各组脑缺血-再灌注损伤后第3,7,14天血清中SDF-1的变化。结果模型对照组SDF-1在不同时间点均有表达,第3天明显增加,第7天达高峰,第14天下降[分别为(545.92±21.81),(574.82±26.58),(558.97±24.17)pg·m L^-1],但仍高于假手术组[分别为(350.09±9.36),(356.62±10.86),(356.62±9.64)pg·m L^-1];通窍活血汤中剂量组和大剂量组血清中SDF-1含量[分别为(560.84±32.37),(655.95±14.87),(625.18±14.77)pg·m L^-1和(595.07±19.19),(760.39±46.90),(679.26±37.01)pg·m L^-1]均高于模型对照组。结论大鼠脑缺血-再灌注会引起SDF-1含量增加,通窍活血汤可促进SDF-1的增加。
Objective To study the expression of stromal cell-derived factor-1( SDF-1) on cerebral ischemiareperfusion injury in rats and the effect of Tongqiao Huoxue decoction. Methods A total of 30 male SD rats were randomly divided into groups a sham operation,a model control and Tongqiao Huoxue decoction at three different doses,6 rats in each group.The model of middle cerebral artery occlusion was set up in rats,the experimental group was perfused with Tongqiao Huoxue decoction,the sham operation and model control groups were treated with distilled water. The changes of SDF-1 were detected on3,7,14 d after ischemia-reperfusion injury by enzyme-linked immunosorbent assay( ELISA). Results SDF-1 in the model control group was expressed at different time point,which enhanced from the third day( 545.92± 21.81) pg·m L^-1,and reached the peak on the 7th day( 574.82±26.58) pg·m L^-1; SDF-1 started to decline on the 14 th days( 558.97±24.17) pg·m L^-1,which were higher than the sham operation group[( 350.09±9.36),( 356.62±10.86),( 356.62±9.64) pg·m L^-1].The content of SDF-1 in the middle-dose and high-dose Tongqiao Huoxue decotion group was( 560.84±32.37),( 655.95± 14.87),( 625.18±14.77) pg·m L^-1and( 595.07±19.19),( 760.39±46.90),( 679.26±37.01) pg·m L^-1,respectively,which were higher than the model control group. Conclusion SDF-1 increases significantly after ischemia-reperfusion injury,and Tongqiao Huoxue decoction could promote the increase.
