目的:利用免疫组化法研究温胆汤、化瘀温胆汤对糖耐量低减( IGT )大鼠脂肪中PI3 K、PKB、GSK-3β蛋白表达的影响。方法:40只SD大鼠随机分为空白对照组、IGT模型组、温胆汤组和化瘀温胆汤组,喂养24周后,取脂肪采用免疫组化法测定PI3 K、PKB、GSK-3β蛋白含量。结果:与空白对照组比较,IGT组PI3K、PKB蛋白含量显著降低(P<0.05),GSK-3β蛋白含量显著升高(P<0.05)。与IGT组比较,温胆汤、化瘀温胆汤均可显著升高PI3 K、PKB蛋白含量( P<0.05),降低GSK-3β蛋白含量( P<0.05)。结论:温胆汤、化瘀温胆汤可以不同程度的作用于IGT大鼠脂肪的PI3K通路。
Objective:The experiment aimed to study the protein expressions of phosphatidyl inositol 3-kinase(PI3K), protein kinase B( PKB) and glycogen synthase kinase-3β( GSK-3β) in the tats′fat by immunohistochemistry method. Methods:The SD rats were divided into four groups at random,which were blank control group,IGT model group,Wen-dan decoction group and Huayu Wendan decoction group.Detect the protein expressions of PI3K,PKB and GSK-3βin the fat by immunohistochemical staining.Results:Compared with the blank control group, the protein expressions of PI3K and PKB in fat of IGT group all decreased,but the protein expression of GSK-3βincreased obviously.Conclusion:Wendan decoction and Huayu Wendan decoction can have an effect on different levels of PI3K pathway in the IGT rats.
