为了给品质改良提供基础材料,并了解西北春麦区小麦地方品种的遗传多样性,采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)方法,分析了493份小麦地方品种的高分子量麦谷蛋白亚基(HMW-GS)的组成。结果表明:在供试材料中,Glu-1位点共有26个等位基因,其中Glu-A1位点3个,Glu-B1位点9个,Glu-D1位点14个,亚基null、7+8、2+12在各自的位点上出现频率最高,分别达到了94.53%、92.92%、86.24%;亚基组成类型共有30种,主要为null/7+8/2+12,频率达79.76%;同时筛选出一些含有1、2*、13+16、14+15、5+10、1.5+10等优质亚基或亚基对的材料,可作为优质基因源;西北春麦区小麦地方品种间Glu-1位点的遗传多样性,以Glu-D1位点最高,其次是Glu-B1位点,Glu-A1位点最低。
In order to provide favorable materials for the quality improvement and detect the genetic diversity of landraces collected from northwest spring wheat production area, subunits composition of high molecular weight gluten with a total of 493 wheat landraees were analyzed by means of SDS-PAGE. The result indicated that there were 26 alleles on Glu-1 loci, including 3 alleles on Glu-A1 loci, 9 alleles on Glu-B1 loci, 14 alleles on Glu-D1 loci; and null, 7 + 8, 2 + 12 subunits occurred with highest frequency on each of loci, reaching 81.36% ,61.92% , 87.52% respectively. There are 30 types of subunit compositions and predominant was null/7 + 8/2 + 12 with a frequency of 79.76%. Materials conferred with elite subunits of 1, 2* , 17 + 18, 14 + 15 and 5 + 10 were selected and could be a source of desirable genes. Among the three locus of Glu-1 loci, Glu-D1 had the highest genetic diversity index, followed by Glu-B1, Glu-A1 had the lowest.
