目的研究亚砷酸钠对人类黑色素瘤细胞凋亡的影响,并比较砷对2种黑色素瘤细胞(G361及A375)凋亡作用的差异。方法2种细胞分别暴露于亚砷酸钠(NaAsO2)0,1,5,10,20,40μmol/L以及促凋亡阳性对照物放射菌素5μg/μl,培养24 h后,收集细胞,用流式细胞仪测定10 000个细胞的凋亡及坏死发生水平。结果2种细胞在20及40μmol/LNaAsO2作用下,凋亡发生水平均明显。高于相应对照组;40μmol/L(G361)及20,40μmol/L(A375)NaA-sO2作用下坏死水平显著高于对照组;暴露于20,40μmol/L NaAsO2以及5μg/μl放射菌素时,A375细胞的凋亡及坏死发生水平均显著低于G361细胞。结论NaAsO2可致人类黑色素瘤细胞发生凋亡,并对2种人类黑色素瘤细胞凋亡率不同。
Objective To observe the effect of sodium arsenite (NaAsO2) exposure on human melanoma cell lines of G361 and A375, and to explore whether there were differences between the effects of apoptosis induced by these two cell lines. Methods Cells were exposed to NaAsO2 of 0, 1,5, 10, 20 and 40 μmol/L, and actinomycin of 5 μg/μl, then collected after being cultured for 24 h. 10000 cells were measured for apoptosis and necrosis rates. Results Apoptosis rate of 20 and 40 μmol/L NaAsO2 groups were significantly higher than that of control in both G361 and A375 ; Necrosis rate of 40 μmol/L NaAsO2 group in G361 and 20, 40 μmol/L NaAsO2 groups in A375 were markedly higher than that of control;apoptosis rate of A375 induced by NaAsO2 exposure of 20, 40 μmol/L and actinomycin of 5 μg/μl were significantly lower than those of G361 .Conclusion NaAsO2 exposure can induce apoptosls in human melanoma cell llne. The effect of apoptosis induced by NaAsO2 is different in different cell lines of human melanoma.
