目的建立胶质瘤血瘤屏障体外模型,研究缓教肽(BK)对胶质瘤血瘤屏障钙激活钾电流(Ikca)的作用,探讨BK开通血瘤屏障的机制。方法条件培养和共培养,膜片钳全细胞记录。结果条件培养的脑微血管内皮细胞(BMECs)形态无明显变化,胶质瘤C6细胞之间相互接触紧密。共培养的BMECs和C6细胞各自聚集,之间形成界面,1μmol·L^-1缓激肽使BMECs和C6细胞界面内钙激活钾电流下降。结论 1μmol·L^-1的缓激肽抑制部分血瘤屏障体外模型BMECs和C6细胞的钙激活钾电流。
Objective To establish the glioma BTB model in vitro to study the effect of bradykinin on Ikca of glioma BTB. Methods Conditioned culture and co-culture, whole cell electrophysiological recording were used. Resuits In the conditioned medium, the brain microvascular endothelial cells (BMECs) were found without morphological change, C6 cells connected tightly. BMECs and C6 cells were aggregated separately and formed a interface between BMCs and C6 cells when the cells were co-cultured. Bradykinin (1 μmol·L^-1 )decreased calcium-activated potassium current of BMECs and C6 cells in the interface. Conclusion Bradykinin inhibits I kca, of C6 BTB.
