目的体外培养兔骨髓基质细胞(Marrow Stromal Cells,MSCs),研究其生物学特性,为骨组织工程应用提供基础。方法抽取新西兰白兔骨髓,进行体外培养并传代,观察细胞生长情况,然后将细胞与牛松质骨(bovine cance Uousbone,BCB)载体复合。通过用倒置显微镜和扫描电镜观察细胞粘附和生长情况,同时测定碱性磷酸酶活性和细胞增殖分化情况。结果骨髓基质细胞在体外培养生长良好,细胞与支架复合后可以在支架表面粘附伸展。结论骨髓基质细胞在体外生长良好,可以作为种子细胞在组织工程中应用。
Objective To investigate biological characteristics of cultured rabbit bone marrow stromal cells and the application of this technique on bone tissue engineering . Methods Bone marrow was cultured and passed to the future generation in vitro after obtaining from healthy rabbits. Cells were then seeded into BCB scaffolds. Their adhesion situation and growth were analyzed by reverse microscope and scanning electron microscope (SME) . The proliferation and differentiation of cells were evaluated by MTT assay and the activity of alkaline phosphatase was detected. Results The data revealed extensive cellular adhesion and grows, little biological behavior of cells was affected. Conclusions MSCs can be cultured by this means and can be used as seeded cell for bone tissue engineering .
