目的:建立准确快速测定大鼠口服葛根后血浆中大豆苷元含量的方法,探讨大豆苷元的体内代谢情况。方法:采用反相高效液相色谱法(RP-HPLC)测定大鼠口服药物后不同时间血浆中大豆苷元的含量,待测血浆样品采用沉淀蛋白法处理,Phenomenex Luna C18高效液相色谱柱(4.60 mm×250 mm,5μm)进行分析,流动相为甲醇-0.1%甲酸水(60:40v/v)。结果:测定方法稳定,给药后5 min血浆中即可检测出大豆苷元,20 min内达到峰值,高、中、中低、低4种浓度的日内精密度均≤5.7%,日间精密度均≤9.3%,回收率分别为89.5%、93.2%、93.9%、84.8%,在0.1~64μg/mL范围内线性关系良好。结论:此方法快捷、灵敏、简便,用于血浆中大豆苷元的含量测定,取得满意结果。
AIM:To explore a RP-HPLC method for determining daidzein content of rat plasma after oral administration of Pueraria lobata.METHODS:The analysis was performed on a Phenomenex Luna C18 column(4.60mm×250 mm,5 μm) with the mobile phase consisted of CH3OH and 0.1% aqueous formic acid(60:40,v/v).The detection wavelength was set at 260 nm.RESULTS:In five minutes after the oral administration,daidzein was detected in rat plasma,and the plasma level reached the peak value within 20 minutes.The calibration curve was linear in the range of 0.1-64 μg/mL.The intraday and inter precisions were less than 5.70% and 9.3%,respectively.The recovery rate of four different concentrations was 89.5%,93.2%,93.9% and 84.8%,respectively.CONCLUSION:In this paper,RP-HPLC proves to be a suitable method for determining the plasma levelof daidzein.
