目的 研究丹参脂溶性成分对大鼠视网膜Müller细胞的毒性作用.方法 以不同浓度的丹参脂溶性成分及丹参酮ⅡA对照品分别干预体外纯化培养的大鼠视网膜Müller细胞48 h,采用CCK-8法检测其细胞增殖抑制率.结果 测得丹参脂溶性成分及丹参酮ⅡA对照品对大鼠视网膜Müller细胞增殖均有一定抑制作用,且具有剂量依赖性.CCK-8法测得丹参脂溶性成分及丹参酮ⅡA对照品的IC50分别为2.181、6.176 μg·mL-1,给药20 μg· mL-1时,测得细胞增殖抑制率分别为75.8% 、58.7%.结论 丹参脂溶性成分及丹参酮ⅡA对照品对大鼠视网膜Müller细胞均有一定毒性,所测IC50值为进一步筛选研究丹参脂溶性成分治疗糖尿病性视网膜病变作用机制的有效浓度提供了一定的实验依据.
OBJECTIVE To study the toxic effect of the liposoluble components of Salvia on rat retinal Mtiller cell. METHODS The rat retinal Muller cell were treated by the liposoluble components of Salvia and tanshinone ⅡA( Tan ⅡA) standard substance under different concentrations for 48 h, the inhibition of cell proliferation of the cell were detected by CCK - 8 method. RESULTS The liposoluble components of Salvia and Tan ⅡA standard substance both showed a certain inhibition effect on rat retinal Mtiller cells proliferation,and the effect depended on the concentration. The ICs0 detected of the liposoluble components of Salvia and Tan HA standard substance were 2. 181 μg·mL-1 and 6. 176 μg·mL-1 by CCK - 8 method, respectively, and the inhibition rates were observed as 75.8% and 58.7% ,respectively,when the concentration was 20 μg· mL-1. CONCLUSION Both of the liposoluble components of Salvia and Tan ⅡA standard substance have a certain toxicity on rat retinal Mtiller cell. The measured ICs0 reveals experimental basis and lais a fundament for the further screening studies of the effective concentration of the liposoluble components of Salvia in the treatment of DR mechanisms.
