目的探讨G蛋白偶联受体激酶结合蛋白1(GIT1)基因表达缺失对骨折愈合过程中软骨细胞增殖和凋亡的影响。方法应用GIT1基因敲除小鼠(A组,30只)和同窝野生型小鼠(B组,30只)制作股骨骨折模型。造模成功后7、14和21d,采用免疫组化和免疫荧光法检测骨痂区软骨细胞增殖能力和凋亡情况。结果与B组相比,A组小鼠骨痂区软骨细胞堆积,软骨细胞增殖能力减弱,凋亡能力下降(P〈0.05)。结论 GIT1基因表达缺失可能会导致骨折愈合延迟。
Objective To investigate the effect of lack of G protein-coupled receptor kinase binding protein 1(GIT1)gene expression on the proliferation and apoptosis of cartilage cells during fracture healing.Methods GIT1-knockout mice(group A,30 mice)and wild-type mice(group B,30mice)were used to establish femoral fracture model.On the 7th,14 th and 21 st day after fracture,the proliferation and apoptosis of cartilage cells in callus area were detected by immunohistochemistry and immunofluorescence methods.Results Compared with group B,cartilage cells were accumulated in callus area and the proliferation and apoptosis of cartilage cells were decreased in group A(P〈0.05).Conclusion Lack of GIT1 gene expression may lead to delayed fracture healing.
