目的研究细胞周期调控蛋白(MRG15)在哺乳动物细胞中的表达与定位。方法以含人MRG15全长cDNA序列的质粒为模板,PCR扩增,构建MRG15真核表达载体,分别利用Western blot法和免疫荧光法检测其在哺乳动物细胞株中的表达与定位。结果 Western blot法检测结果表明MRG15在哺乳动物细胞中能有效地表达,免疫荧光法结果显示MRG15主要分布在细胞核。结论 MRG15蛋白主要在细胞核中表达,为进一步了解MRG15的功能奠定了一定基础。
Objective To investigate the location and expression of the cell cycle regulatory protein(MRG15) in mammalian cells.Methods The full length cDNA sequence of MRG15 from plasmid was used to amplify with PCR,to conduct the eukaryotic expression vector of MRG15.Immunofluorescence and Western blot were adopted to detect the location and expression of MRG15 in the cell strain.Results Western blot results showed effective expression in the cells.Besides,the results of immunofluorescence indicated that MRG15 was located in cell nucleus.Conclusion The expression of MRG15 protein is localized in the nucleus,which lays foundation to further study of the function of MRG15.
