目的研究细胞内氯离子通道蛋白2(CLIC2)在细胞中的定位与表达。方法以含人CLIC2全长cDNA序列的菌液为模板,PCR法构建CLIC2真核表达载体,利用免疫荧光和Western blot法检测其在细胞株中的定位与表达。结果细胞免疫荧光结果显示CLIC2在细胞质、细胞核中均有分布;Western blot的结果证明其在细胞中能有效地表达。结论CLIC2蛋白在细胞质和细胞核中均大量分布,为进一步了解CLIC2的功能奠定了基础。
Objective To investigate the location and expression of the chloride intracellular channel 2 (CLIC2) in human cells. Methods Bacteria liquid containing the full-length human cDNA sequence of CLIC2 was used as PCR template to construct the eukaryotic expression vector. Immunofluorescence and Western blot were adopted to detect the location and expression of CLIC2 in the cell strain. Results The results of immunofluorescence indicated that CLIC2 was located in both cytoplasm and cell nucleus. Besides, Western blot results showed that it can be effectively expressed in the cells. Conclusion CLIC2 is widely distributed and expressed in human cells, which lays foundation to further study the function of CLIC2.
