目的探讨人参皂苷Rh2对人白血病HL-60细胞增殖和凋亡的影响。方法应用MTT法检测人参皂苷Rh2对HL-60细胞增殖的影响,采用DNA的片段化实验,观察人参皂苷Rh2对HL-60细胞凋亡的影响。结果G—Rh2能明显抑制HL-60细胞增殖,呈浓度依赖性关系;人参皂苷Rh2能明显诱导HL-60细胞凋亡,呈浓度和时间依赖效应,DNA的琼脂糖凝胶电泳分析可见凋亡特征性的DNA梯状图谱。结论G—Rh2主要通过有效抑制HL-60细胞增殖并诱导其凋亡从而发挥其抗白血病效应。
[Objective] To investigate effect of Ginsenoside Rh2 on the proliferation and apoptosis in HL-60 cells. [Method] Effects of Ginsenoside Rh2 on the HL-60 ceils proliferation and apoptosis were determined by MTT [3-(4, 5--dimethylthiazol-2-yl)-2, 5-diphenyl tetrasodium bromide] and DNA fragmentation assays, respectively. [Resultsl Ginsenoside Rh2 can significantly inhibit the proliferation of HL-60 ceils in the dose--dependent manner. In addition, apoptosis of HL-60 cells can be induced by Ginsenoside Rh2 ip the dose- and time-dependent manners. Apoptosis-characteristie DNA ladders can be observed by DNA agarose gel electrophoresis. [Conclusions] Ginsenoside Rh2 plays an crucially important role in exerting its anti-leukemic effects via inhibition of proliferation and induction of apoptosis in HL-60 ceils.
