为筛选太行鸡脾脏组织中绵羊红细胞(sheepredbloodcell,SRBC)免疫应答的关键基因和信号通路,本研究采用高通量测序技术,构建了太行鸡SRBC免疫组和对照组脾脏组织的数字基因表达谱,对差异表达的基因进行筛选与注释、GO分析和通路的功能显著性富集以及差异基因编码蛋白的互作网络分析。结果表明:太行鸡在免疫6d后的SRBC抗体滴度平均值为7.653;与对照组相比,差异倍数在2倍以上的共有1108个基因,其中496个上调表达,612个下调表达,412个上调基因只在免疫组表达,537个下调基因仅在对照组表达;上调基因主要涉及T细胞激活、淋巴细胞增殖和单核细胞增殖等生物学过程,下调基因主要参与免疫效应、防御效应、炎性效应、先天性免疫效应生物学过程;在分子注释系统数据库中注释到的免疫相关的显著调控通路有12条,包括抗原的加工递呈、Toll样受体信号通路、沙门氏菌感染、HTLV—I感染和单纯疱疹病毒感染等;56个差异基因编码的蛋白产物存在相互作用,其中16个基因位于网络中心节点,部分差异基因与SRBC相关QTL重叠。初步认为抗原加工与递呈的MHCII类分子途径、T细胞激活等调控通路及RAG2、FOS、MHC B-LBII、HSP90AAl、CXCR4和HSPA8等基因可能是参与调控太行鸡SRBC免疫应答的重要信号通路和基因,但其功能和影响还有待于深入研究。
In order to find the immune response genes and key pathways associated with sheep red blood cells in the spleen of Taihang chicken.The digital gene expression (DGE) profile in the spleen of Taihang chicken SRBC immunized group and control group were built by high throughout sequencing technology, the different expression genes were screened and annotated. Significant enrichment analysis of GO and pathway were done. The protein interactions network analysis for the differentially expressed genes was also built by STRING soft . The results showed that the average anti-SRBC antibody titer was 7.653 after 6 days immunized with 1 mL SRBC. SRBC immunized group compared to control group, different multiples more than two times were 1 108, among them, 496 were up regulated genes and 612 were down regulated genes, 412 genes expressed only in SRBC immunized group, 537 genes expressed only in control group. GO analysis indicated that the up regulated genes mainly involved in biological processes including T cell activation,leukocyte proliferation and mononuclear cell proliferation, and the down regulated genes mainly involved in biological processes including immune response, defense response , inflammatory response innate immune response.The pathway analysis suggested that the differently expressed genes mainly took part in antigen processing and presentation, Toll- like receptor signaling pathway, salmonella infection, HTLV-I infection and herpes simplex infection pathways. The protein products of 56 differentially expressed genes in spleen had interactions, 16 of them located in the center node of interaction network. Some differentially expressed genes were found to be overlapped with the QTLs associated SRBC traits.
