为了研究鸡传染性支气管炎病毒弱毒株(IBV ZZ-V35)在鸡胚中的增殖规律,用尿囊腔接种法给11日龄的SPF鸡胚接种IBV ZZ-V35弱毒,并构建了阳性质粒,采用Taq Man探针实时定量PCR(real-time PCR)法建立了标准曲线,分别检测感染鸡胚尿囊液中36、48、52、56、60、64、68、72、76、80、88、96、120、144h的病毒含量。结果显示,在鸡胚发育不同的时间点IBV ZZ-V35弱毒的拷贝数分别为1.416×106、1.791×106、2.056×106、3.027×106、3.664×106、1.854×106、1.380×106、1.109×106、7.976×105、5.408×105、2.958×105、2.512×105、4.365×104、1.130×104。研究发现,IBV ZZ-V35弱毒的拷贝数在60h达到最高值,即3.664×106。这为IBV ZZ-V35弱毒株疫苗的研究提供了新的依据。
To explore the proliferative rule of infectious bronchitis virus (IBV) attenuated strain, IBV ZZ- V3swas inoculated in the allantoic cavity of the l 1-day old SPF chicken embryo. Real-time PCR based on TaqMan probe was used to test the proliferative rule of IBV in the allantoic cavity. After the inoculation of IBV ZZ-V3s in 11-day old SPF chicken embryoes,the real-time PCR was used to detected allantoic fluid in 36,48,52,60,68,72,76,80,88,96,120,144 h. The results showed that the viral copies are 1. 416 × 106, 1. 791×106,2. 056×106,3. 027×106,3. 664×106,1. 854×106,1. 380×106,1. 109×106,7. 976×105, 5. 408 ×105 , 2. 958 × 105 , 2. 512 × 105 , 4. 365 × 104 and 1. 130 × 104 , respectively. It showed that the maximal copies occured in 60 h after inoculation, which provided new theoretical basis for the research of vaccine of IBV ZZ-V35.
