目的分析去甲斑蝥素(NCTD)作用于肝癌细胞SMMC-7721前后蛋白质组表达的差异。方法采用四氮唑蓝还原法(MTT)测定细胞生长抑制率。20μg/ml的NCTD作用于肝癌细胞SMMC-772124h,分别收集处理组与对照组细胞,裂解并提取细胞内总蛋白;双向凝胶电泳(2-DE)分离蛋白质,凝胶银染,ImageMaster2D Platinum软件分析2-DE图谱,寻找差异表达蛋白点。结果NCTD能诱导肝癌细胞SMMC-7721的凋亡,具有时间和剂量依赖性,与对照组比较,处理组2-DE图谱中有5个蛋白点表达上调,6个蛋白点表达下调。结论抗癌药NCTD能诱导肝癌细胞SMMC-7721凋亡并引起细胞内蛋白质组表达的改变。
Objective To analyze differential proteome of SMMC-7721 cells treated with norcantharidin(NCTD) by twodimensional gel electrophoresis(2-DE). Methods Inhibitory effects of NCTD on SMMC-7721 cells were measured by MTF test. SMMC-7721 cells were exposed to NCTD (20 μg/ml) for 24 hours. The control and treated cells were collected respectively. Total proteins were extracted using lysis buffer and separated by 2-DE. The gels were stained with sliver. Different expression proteins were analyzed with ImageMaster 2D Platinum software. Results NCTD induced SMMC-7721 cell death in a time and dose dependent manner. Compared with the control ,five protein spots were identified to be up-regulated and six down-regulated in the treated group. Conclusion The anti-tumor drug NCTD can induce the apoptosis of SMMC-7721 cells,and alter their proteomic expression.
