目的探讨糖尿病高血压大鼠肾脏单核细胞趋化蛋白1(MCP-1)表达变化及舒洛地特对MCP-1表达的影响。方法18只Wistar大鼠随机分为3组:对照组(C组)、糖尿病高血压组(D组)、舒洛地特治疗组(s组),每组各6只。采用链脲佐菌素(STZ)+醋酸脱氧皮质酮(IX)CA)+盐水建立糖尿病高血压大鼠模型。第8周测定尾动脉压、尿白蛋白。于第8周末处死动物,PAS染色观察肾小球病理改变,免疫组织化学法及逆转录聚合酶链反应(RT-PCR)分别检测MCP-1蛋白及mR—NA表达改变。结果第8周末D组尿白蛋白量及肾组织MCP-1表达显著高于C组(P〈0.05),S组尿白蛋白量及肾组织MCP-1表达较D组降低(P〈0.05)。结论舒洛地特可通过降低。肾组织MCP-1表达,减轻糖尿病高血压大鼠病理损伤。
Objective To investigate the effects of sulodexide on the expression of monocyte cbemoattractant protein (MCP-1) in streptozotocin (STZ) diabetic desoxycortieosterone acetate (DOCA)-hypertensive rats. Methods Wistar rats were randomly divided into 3 groups: control group (C), STZ diabetic IX)CA-hypertensive group(D) and sulodexide-treated group(S). Systolic pressure and urinary albumin for 24 h were measured: Animals were sacrificed at the week 8. Renal morphological changes were examined under the light microscopy. The expression of MCP-1 in kidney was detected by immunohistochemistry and RT-PCR. Results The expression of MCP-1 in kidney was increased remarkably in STZ diabetic DOCA-hypertensive group as compared with control group(P%0. (15). As compared with STZ diabetic DOCA-hypertens~ve group, the expression of MCP-1 was significantly de- creased in sulodexide-treated group(P〈0. 05). Conclusions Sulodexide exerted renoproteetive effects i'n STZ diabetic IX)CA-hypertensive model. The mechanism may be at least partly correlated with the suppression of increasing expression of MCP-1 in kidney.
