目的:研究替米沙坦对醋酸去氧皮质酮(DOCA)高血压大鼠肾脏保护作用和足细胞Podocalyxin蛋白表达的影响。方法:18只雄性Wistar大鼠随机分为对照组(A组)、高血压组(B组)和替米沙坦治疗组(C组)。单肾切除后皮下注射DOCA(5 mg/只,2次/周)和饮用盐水(1%Nacl+0.2%KCl)建立高血压模型。C组用替米沙坦[5mg/(kg.d)]灌胃。测定第4、8周尿蛋白。8周后处死动物,测血肌酐(SCr)、血钠、血钾,取肾组织PAS染色观察病理变化,免疫组化法检测肾脏podocalyxin表达变化。结果:B组4周始血压和尿蛋白较A组逐渐升高(P〈0.05)。B组肾小球硬化指数和间质纤维化指数均明显高于A组(P〈0.05),podocalyxin蛋白表达较A组降低。C组血压、尿蛋白和病理损伤较B组减轻,podocalyxin表达显著高于B组(P〈0.05)。结论:替米沙坦通过上调足细胞podocalyxin表达,减轻DOCA高血压大鼠肾损伤。
Objective: To investigate the effect of telmisartan on the renal function and podocalyxin ex- pression in podoeytes of deoxycorticosterone acetate (DOCA) hypertensive Wistar rats. Methods: Eighteen male Wistar rats were randomly assigned into three groups: control group, hypertensive group, and telmisartan-treated group (n= 6 respectively). Blood pressure and proteinuria were measured after 4 and 8 weeks. Renal pathological changes were quantified by the indices of glo- merulosclerosis (GS) and interstitial fibrosis (IF). Expression of podocalyxin was analyzed by immunohistochemical staining. Results: The proteinuria and hypertension in hypertensive rats were significantly alleviated after telmisart~ n treatment. In addition, down-regulation of podoca- lyxin expression in glomeruli and pathological damages in hypertensive rats were prevented by telmisartan intervention. Conclusion: Telmisartan exerts a protective effect on improving expres- sion of podocalyxin in glomeruli to prevent the progression of hypertensive renal damage.
