目的:研究表没食子儿茶素没食子酸酯(epigallocateehin-3-galiate,EGCG)对舌鳞癌细胞CAL-27的表皮生长因子受体(epidermal growth factor receptor,EGFR)、相对分子质量为67000的层粘连蛋白受体(67000 dalton laminin receptor,67LR)及血管内皮生长因子(vascula rendothelial growth factor receptor,VEGF)表达的影响,从而揭示EGCG抑致舌鳞癌细胞增殖的可能机制。方法:不同浓度EGCG处理CAL-27细胞24h后.MTT法检测细胞增殖活性:RealTime PCR法和Western Blot法分别检测CAL-27细胞EGFR、67LR、VEGF的mRNA及蛋白表达情况;Western Blot法检测磷酸化EGFR(p-EGFR)的蛋白表达水平。结果:EGCG浓度依赖性抑制CAL.27细胞的增殖,且抑制其EGFR、67LR、VEGF的mRNA和蛋白、P—EGFR的蛋白表达。结论:EGCG能影响EGFR、67LR及VEGF的转录和翻译水平以及D—EGFR蛋白表达.这可能是EGCG抑制舌鳞癌细胞CAL-27增殖的重要机制。
Objective: This study was designed to detect the effect of epigallocatechin-3-gallate (EGCG) on the expression of EGFR, 67LR and VEGF in human tongue squamous cell carcinoma cell line CAL-27, and the role of proliferation inhibition to cancer cells. Methods: Human tongue squamous eell carcinoma cell line CAL-27 were treated with EGCG for 24h. Cell viability was assessed by MTY assay. The mRNA levels of EGFR, 67LR and VEGF were measured by real- time PCR and their protein levels were determined by Western Blot. The protein expression of p-EGFR were evaluated by Western Blot. Results: EGCG showed inhibition effect on CAL-27 ceils proliferation. Suppression effect of the mRNA levels and protein levels of EGFR, 67LR and VEGF, protein levels of p-EGFR were manifested in a close-dependent manner. Conclusion: EGCG inhibited the mRNA and protein expression of EGFR, 67LR and VEGF, as well as protein levels of p-EGFR, which may be associated with its suppression effect of proliferation of CAL-27 cells.
