目的研究表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG)及镍离子(Ni2+)共同作用对舌鳞癌细胞系Cal-27 cyclinD1和survivin基因表达的影响。方法 MTT法检测Cal-27的细胞存活率;real-time PCR法检测cyclinD1及survivin mRNA水平表达的影响;Western blot法检测cyclinD1和survivin蛋白水平的表达情况。结果高浓度EGCG(〉100μmol/L)可明显抑制Cal-27生长,且对细胞cyclinD1和survivin基因转录及蛋白表达有明显下调作用(P〈0.05);Ni2+与EGCG共同作用增加了对Cal-27的抑制作用,cyclinD1和survivin表达更加显著下降(P〈0.05)。结论 Ni2+与EGCG共同作用后可增强EGCG对细胞生存率及cyclinD1和survivin基因表达的抑制作用,两者起协同作用。
Objective To determine the effect of epigallocatechin gallate(EGCG) and Ni2+ on the expression of cyclinD1 and survivin in tongue squamous carcinoma cells(Cal-27). Methods The viability of cells was determined by the MTT reduction assay. Real-time PCR was used to examine the mRNA expression of cyclinD1 and survivin. Western blot method was used to analyze the expression level of protein of cyclinDl and sl~rvivin. Results The proliferation of Cal-27 cells was significantly inhibited by high concentration of EGCG (greater than 100 μmol/L). The mRNA expression and protein expression of cyclinD1 and survivin were also downregulated (P〈0. 05). Ni2+ could enhance the inhibition of Cal-27 cells by EGCG. The expression of cyclinDl and survivill were significantly suppressed by EGCG plus Ni2+ (P〈0.05). Conclusions The inhibition of Cal-27 cells and expression of cyclinDl and survivin by EGCG could be enhanced by the mixture of Ni2+ .
