目的:评价可注射纳米羟基磷灰石/半水硫酸钙(n HAC/CSH)植骨材料的细胞相容性。方法:犬外周血基质细胞(d PBSCs)从健康成年犬的外周血中分离得到,通过直接接触或浸提液法检测n HAC/CSH的细胞相容性。制备n HAC/CSH的浸提液,通过MTT法检测细胞的增殖,采用流式细胞仪评价材料对细胞凋亡的影响,n HAC/CSH与d PBSCs共培养后光镜和扫描电镜下观察细胞形态。结果:随着培养时间的延长,对照组和浸提液组细胞数量不断增加,3天时两组细胞的凋亡比率相近,n HAC/CSH和d PBSCs共培养时,细胞伸展良好。结论:n HAC/CSH具有良好的细胞相容性。
Objective: The aim of the study was to assess the cell biocompatibility of injectable nano-hydroxyapatite/collagen/calcium sulphate hemihydrate(n HAC/CSH) bone cement. Methods: Dog peripheral blood stromal cells(d PBSCs) were isolated from peripheral blood of healthy adult dogs. An in vitro study on the cytocompatability of n HAC/CSH was conducted. The d BMPCs morphology, proliferation and apoptosis assays were conducted using the direct contact and extraction method by microscopic observation, MTT analysis and flow cytometric method. Results: The MTT assay showed that proliferation proceeded in two groups. There was a spontaneous apoptosis of d BMPCs, with similar apoptosis rates on Day 3 in both the control culture and the extract group. Cells grew with n HAC/CSH cement readily attached to it and exhibited an elongated appearance. Conclusion: Studies in vitro revealed that the n HAC/CSH cement has good cell biocompatibility.
