Hes属于bHLH(碱性螺旋-环-螺旋)转录因子,具有bHLH和Orange两个结构域、保守四肽WRPW。目前研究认为Hes1、Hes5、Hes7是Notch信号通路的下游靶基因,而Hes2、Hes3、Hes6似乎不依赖于Notch信号通路。Hes在发育决定(如神经发育、体节发育、骨骼发育、T淋巴细胞发育、癌症发生等过程)中起着很重要的作用。研究Hes所参与的发育调控网络及其作用机制,必将对我们更好的理解它们的生物学功能和应用于临床治疗提供指导作用。鉴于此,本文综述了Hes在发育决定过程中及在癌症发生过程中的作用及其研究进展。
Hes genes,which are the members of basic helix-loop-helix(bHLH) transcription factors,contain bHLH domain,Orange domain and WRPW tetrapeptide.Recent studies revealed that Hes1,Hes5 and Hes7 were target genes of Notch signaling pathway.However,Hes2,Hes3 and Hes6 appeared to be independent of Notch signaling pathway.Hes genes play essential roles in developmental decisions,such as nervous system development,somite segmentation,skeleton development,T lymphocytes development and cancer.Studies of the physiology characters and mechanisms of Hes-mediated developments will give some clues on understanding of their biological roles and benefit to the clinical diagnosis and therapy.In order to understand the roles of Hes genes,we summarized the research proceedings of Hes genes in development and cancer.
