目的:探讨不同剂量N-甲基-D-天冬氨酸受体拮抗剂MK-801对利血平诱导的行为性抑郁大鼠抑郁样行为的影响。方法:SD雄性大鼠(40只)随机分为正常对照组,利血平(4mg/kg)组,MK-801(0.1、0.3、0.5mg/kg)+利血平(4mg/kg)组,每组8只。采用开场实验和强迫游泳实验检测大鼠的抑郁样行为。结果:腹腔注射利血平(4mg/kg)1 h后,开场实验示利血平组总路程、中央活动路程较正常对照组显著减少(P〈0.01),中央活动时间也较正常对照组减少(P〈0.05)。MK-801+利血平组总路程均较利血平组显著增多(P〈0.01),0.3、0.5mg/kgMK-801+利血平组中央活动路程和时间均较利血平组增多(P〈0.05)。0.3、0.5mg/kgMK-801+利血平组中央活动路程较0.1mg/kgMK-801+利血平组增多(P〈0.05)。开场实验中,0.5mg/kgMK-801+利血平组出现共济失调不良反应;强迫游泳实验示利血平组不动时间较正常对照组显著增多(P〈0.01)。MK-801+利血平组不动时间较利血平组显著减少(P〈0.01)。结论:提前给予MK-801可拮抗腹腔注射利血平(4mg/kg)诱导的行为性抑郁,这种效应与MK-801的剂量有一定关系。本实验中0.3mg/kgMK-801是一个较合适的剂量。
Objective:To investigate the behavioral effects of different dose of N-methyl-D-aspartate receptor antagonist MK-801 in reserpine-induced behavioral depressive rats.Methods:Fourty male SD rats were randomly divided into control group,reserpine(4 mg/kg)group,MK-801(0.1、0.3、0.5 mg/kg) + reserpine(4 mg/kg)group.The depressive-like behaviors were checked by open field test and forced swim test.Results:One hour after intraperitoneal injection of reserpine(4 mg/kg),the open field test showed that the total distance and the distance travelled in central zone of reserpine group were significantly decreased compared to the control group(P0.01),the time travelled in central zone was decreased(P0.05).The total distance of MK-801+reserpine group was significantly increased compared to the reserpine group(P0.01).The distance and time travelled in central zone of 0.3、0.5 mg/kg MK-801 + reserpine group were increased compared to the reserpine group(P0.05).The distance travelled in central zone of 0.3、0.5 mg/kg MK-801+reserpine group was increased compared to the 0.1 mg/kg MK-801 + reserpine group(P0.05).The ataxia adverse effect was observed in 0.5 mg/kg MK-801 + reserpine group in open field test.The forced swim test showed that the immobility time of reserpine group was significantly increased compared to the control group(P0.01),the immobility time of MK-801+ reserpine group was significantly decreased compared to the reserpine group(P0.01).Conclusion:MK-801 can antagonize the behavioral depression caused by intraperitoneal injection of reserpine(4 mg/kg),and 0.3 mg/kg MK-801 is a suitable dose in this test.
