目的:研究利血平导致的行为性抑郁大鼠海马神经细胞凋亡与神经元数量的改变。方法:腹腔注射利血平(4mg/kg)复制大鼠的行为性抑郁模型,用自发活动检测大鼠的行为性抑郁,以TUNEL法检测海马神经元凋亡,用尼氏染色观察海马神经元数量。结果:腹腔注射利血平48h后大鼠出现行为性抑郁。利血平可导致大鼠海马CA1区(P〈0.01)、CA3区(P〈0.05)和齿状回神经元(P〈0.001)数量减少,同时检测到此三个区域神经元凋亡的增加(CA1区P〈0.01,CA3区P〈0.001,齿状回区P〈0.001)。结论:利血平诱导的行为性抑郁大鼠可出现海马神经元数量减少,海马神经元凋亡增加是其原因之一。
Objective: To investigate the neuron number and apoptosis in hippocampus in rats treated with reserpine. Methods: Reserpine was injected(i, p. )in rats and the behaviors were checked with locomotor activity. Niss's staining was used to observe the numbers of hippocapal neurons, and TUNEL was used to investigate apoptosis of hippocampal neurons. Results: The behavioral depresssion could be caused at 48 hours after intrapefitoneal injection of reserpine(4 mg/kg). Reserpine decreased the number of neurons in hippocampal in CAI( P 〈 0.01), CA3( P 〈 0.05)and dentate gyms( P 〈 0.001), while the neuronal apoptosis increased in these three areas(CA1 region P 〈 0.01, CA3 region P 〈 0.001, dentate gyrus region P 〈 0.001). Conclusion: The number of hippocampal neurons decreases and the apoptosis increases in the behavioral depressed rats induced by reserpine.
