目的研究熊果酸(UA)对舌鳞癌细胞增殖与凋亡的作用及其可能机制。方法人舌鳞癌细胞Tca8113分别加入UA 0、10、20、40μmol/L,培养12-72h,采用MTT法检测细胞增殖,Hoechst 33342染色观察细胞凋亡的形态学变化,Western blot法检测药物作用后细胞中环氧化酶2(COX-2)、B细胞淋巴瘤-白血病2(Bcl-2)、Bcl相关X蛋白(Bax)及半胱天冬氨酸蛋白酶3(Caspase-3)的蛋白表达。结果 UA呈剂量依赖性地抑制舌鳞癌细胞Tca8113增殖,诱导舌鳞癌细胞凋亡,下调Tca8113细胞中COX-2蛋白表达,上调Bax/Bcl-2比值及活化的Caspase-3蛋白表达。结论 UA可能通过下调COX-2的表达抑制舌鳞癌细胞增殖,诱导凋亡。
Objective To investigate the regulatory effect and underlying mechanism of ursolic acid(UA) on the proliferation and apoptosis of oral tongue squamous carcinoma cells.Methods Oral tongue squamous carcinoma cell Tca8113 was incubated with UA 0,10,20 and 40 μmol/L for 12-72 h.The cell proliferation and morphologic changes of cell apoptosis were determined by MTT assay and Hoechst 33342 staining,respectively.The protein expressions of COX-2,Bcl-2,Bax and Caspase-3 were detected by Western blot.Results UA significantly inhibited the proliferation of Tca8113 cells,induced cell apoptosis,downregulated protein expression of COX-2,and upregulated Bax/Bcl-2 ratio and protein expression of Caspase-3 in a dose-dependent manner.Conclusion UA inhibits proliferation and induces apoptosis of Tca8113 cells probably via downregulating COX-2 expression.
