利用生物素-亲和素系统,以IAsys生物传感器为研究方法,构建了一个实时监测sIL-1R I的实验体系.在生物素表面的样品池结合亲和素,并将生物素衍生化的IL-1α固定于样品池表面,继而将sIL-1R I加入样品池中,通过样品池再生,实现了对sIL-1R I的连续监测.每一步结合反应后,均用PBS/T冲洗以去除非特异性吸附.实验结果表明:生物素衍生化的IL-1α以0.42ng/mm^2的密度被有效地固定于生物素包被的样品池表面.sIL-1R I与固定在样品池表面的IL-1α发生特异性亲和反应,其响应值与sIL-1R I浓度成线性关系,相关系数为0.9913,并且牛血清白蛋白作为sIL-1R I 的保护剂对检测信号没有干扰作用.说明该实验体系可对sIL-1R I进行浓度依赖性、特异性的快速检测.
A method to monitor type 1 soluble interleukin-1 receptor (sIL-1R I ) was developed using the IAsys biosensor together with the avidin-biotin system in real time. First avidin was immobilized on the sensing surface of the biotin cuvette, then biotinylated interleukin-1α (IL-1α) was immobilized on the sensing surface of the avidin-coated biotin cuvette, and sIL-1R I was injected to the IL-1α -coated biotin cuvette. The interaction of sIL-1R I with IL-1α was monitored in real time. At the end of each association step, the reaction cuvette was washed with PBS/T to dissociate the nonspecific association. The experimental results show that IL-1α is immobilized on the surface of cuvette based on biotin without loss of biological activity with the density of 0.42 ng/mm^2. sIL-1R I then binds to immobilized IL-1α specifically in a dose-dependent manner with the correlation coefficient of 0.9913. In addition, bovine serum albumin, the protector of sIL-1R I, does not produce the interferential biosensor signal change for the monitoring of sIL-1R I using the IAsys biosensor based on IL-1α. IAsys biosensor provides an effective assay for real time monitoring sIL-1R I with simple and quick operation.
