以河南教育学院小麦育种研究中心培育的小麦新品系为供试材料,选用C17为基本培养基,分别研究了低温预处理1、2、3、4、5、6、7 d,添加0(CK)、80、100、120 mg/L的聚乙二醇(PEG)对小麦花药出愈率的影响。结果表明:低温预处理4 d的小麦花药出愈率最高(2.108%),且稳定性较好(变异系数0.067),与低温预处理2 d的小麦花药出愈率差异显著(P〈0.05),与其他5个处理之间的差异极显著(P〈0.01)。在培养基中加入80、100、120 mg/L PEG可显著提高小麦花药出愈率,尤其以100 mg/L PEG效果最佳。
Taking the wheat lines bred by Henan Institute of Education as tested materials and C17 as the basic culture medium,the influence of different low temperature pretreatment times and different PEG concentrations added in the medium on wheat anther callus rate was studied. The results showed that the callus rate of wheat anther was the highest(2. 108% ) when it was pretreated by low temperature for 4 d, which had good stability(CV = 0. 067). On the other hand,adding 80—120 mg / L of PEG in the medium could significantly increase the callus rate of wheat anther,the effect of 100 mg / L PEG being the best.
