‘绿波’核桃叶片愈伤组织的诱导研究

本研究以"绿波"核桃试管苗幼嫩叶片为外植体,探讨暗培养时间、植物生长调节剂种类及配比等对愈伤组织的诱导影响。结果表明,暗培养时间对叶片愈伤组织的形成影响较大,暗培养2周时叶片愈伤率可达100%,暗培养时间的增加或减少会抑制叶片愈伤组织的形成,使愈伤组织变得疏松;低浓度的TDZ(2.0~4.0 mg/L)促进叶片愈伤的形成,高浓度的TDZ(8.0~10.0 mg/L)抑制叶片愈伤的形成,同时也抑制了叶片和愈伤组织的褐化,使愈伤组织的结构也变得更加疏松;低浓度的IBA(0.01~0.5 mg/L)有利于叶片愈伤组织的形成,高浓度的IBA(1.0 mg/L)不利于愈伤组织的形成。本研究结果可进一步明确影响核桃叶片愈伤组织诱导的主要因素,为容易分化出不定芽的愈伤组织的诱导研究指明方向。

英文摘要:

In vitro leaves of Juglans regia cv.‘Lvbo'walnut were used as explants, and the effects of dark culture,types and ratio of plant growth regulators(PGRs) on calli induction were studied. The results were as follows. It is more favorable to leaves calli induction for DKW medium. There was a significant influence on leaves calli formation for dark incubation time. After 2 weeks of dark, call genesis rate of leaves was up to 100%. The increase and reduce of dark incubation time would inhibit the formation of leaves calli and made the structure of calli porous. Low concentration of TDZ(2.0 mg/L to 4.0 mg/L) promoted the formation of leaves calli, but the high concentration of TDZ(8.0 mg/L to 10.0 mg/L) inhibited the formation and browning of leaves calli and made the structure of calli more porous. Low concentration of IBA(0.01 mg/L to 0.5 mg/L) was conducive to leaves calli formation, and the high(1.0 mg/L) was bad for leaves calli formation. This study further defined the main factors influencing the walnut leaf calli induction, and gave a research direction for calli induction easy to differentiate into the adventitious bud.
