目的 探讨铝对大鼠肾上腺嗜铬细胞瘤(PC12)细胞络丝蛋白(Reelin)蛋白及基因表达的影响。方法 将处于对数生长期的PC12细胞分别暴露于含终浓度分别为0(对照组)、50、100、200和400μmol/L麦芽酚铝[Al(mal)3]的培养基中,分别染毒12、24和48 h。分别采用酶联免疫吸附测定法(ELISA)、荧光实时定量聚合酶链反应(qRT-PCR)测定Reelin蛋白含量及基因表达。结果 200、400μmol/L Al(mal)3染毒PC12细胞12、24和48 h,Reelin mRNA和Reelin蛋白表达较对照组明显降低(P〈0.05)。结论 Al(mal)3染毒可导致PC12细胞Reelin表达下降。
Objective To investigate the effect of aluminum on mRNA and protein expression of Reelin in rat pheochromoeytoma (PC12) cells. Methods The PC12 cells at the logarithmic growth were exposed to the medium, including aluminum maltolate [Al(mal)3] at the final concentrations of 0 (control group), 50, 100, 200 and 400 μmol/L for 12 h, 24 h and 48 h, respectively. The protein and mRNA expression of Reelin were measured by enzyme linked immunosorbent assay (ELISA) and quantitative real-time polymerase chain reaction (qRT-PCR). Results The mRNA and protein expression of Reelin in PC12 cells exposed to 200 and 400 μmol Al(mal)3 at 12 h, 24 h and 48 h were significantly lower than thosein the control group (P〈0.05). Conclusion Al(mal)3 exposure may decrease the expression of Reelin in PC12 cells.
