Notch family is a series of highly conserved proteins. Notch regulates a broad spectrum of cell development and differentiation. More and more recent studies have addressed the role of Notch signaling on immunity by regulating multiple immune cells. This article reviewed the composition of Notch family, its regulatory factors and target genes, the regulation of Notch signaling on haematopoietic stem ceils, innate immune and adaptive immune related immune cells. In conclusion, Notch signaling play very important roles in the development and functioning of haematopoietic stem ceils, macrophages, dendritic cells, mast cells, T and B lymphocytes, as well as the genesis of immune related diseases including cancer,virus infection, inflammation and autoimmune diseases.
