目的观察低频超声辐照联合微泡对体外培养的大鼠主动脉平滑肌细胞凋亡的影响。方法联合不同浓度的SonoVue(0、0.5×106、1.0×106、1.5×106、2.0×106/mL),使用42.6kHz超声分别辐照大鼠主动脉平滑肌细胞10s、20s、30s,空白对照组不作任何处理。台盼蓝染色分析辐照后即刻细胞存活率,流氏细胞仪观察辐照后24h细胞凋亡率。结果细胞的死亡和凋亡与微泡浓度及辐照时间相关。当超声波在微泡浓度达到或超过0.5×106/mL,联合辐照30s,以及微泡浓度达到2.0×106/mL,联合辐照10s、20s时,细胞大量死亡。在超声辐照10s时,辐照后24h细胞凋亡率随微泡浓度的增加而增加,但在微泡浓度低于1.0×106/mL时,细胞即刻存活率并无显著减少。在超声辐照20s时,细胞即刻存活率随微泡浓度的增加而减少,辐照后24h细胞凋亡率在微泡浓度为0.5×106/mL、1.0×106/mL、1.5×106/mL时差异无统计学意义。结论在一定空化强度范围内,低频超声辐照后24h细胞凋亡率随强度增加而增加,但超过一定程度,则无法增加细胞凋亡率,只会导致细胞死亡。低频超声辐照联合微泡的空化作用可在短时间...
