目的研究糖肾方对糖尿病肾病大鼠的保护作用。方法50只Wistar大鼠随机分为空白组、模型组、蒙诺组(0.83mg/kg)、糖肾方大剂量组(2.67g/kg)、糖肾方小剂量组(1.33g/kg),每组10只。利用单侧肾切除加腹腔注射链脲佐菌素造成糖尿病肾病大鼠模型,连续给药20周。动态检测体重、血糖和尿蛋白/肌酐水平。实验结束时腹主动脉取血检测血浆黏度、血清总蛋白、白蛋白、总胆固醇、甘油三酯、血尿素氮和肌酐水平,取肾组织进行病理组织学观察并对损害程度进行评分。结果与模型组相比,糖肾方小剂量组体重从第4周起显著升高(P〈0.05),从第12周起差异更为显著(P〈0.01)。糖肾方大剂量能显著降低模型动物尿蛋白/肌酐(P〈0.05)、血浆黏度(P〈0.05)、胆固醇(P〈0.05)及甘油三酯含量(P〈0.01),降低肾小球硬化指数(P〈0.01)和肾小管间质纤维化指数(P〈0.01),效果优于蒙诺组。结论糖肾方对糖尿病肾病大鼠具有良好的保护作用,其作用机制可能与改善血液流变学和血脂代谢紊乱有关。
Objective To study the renal protective effect of Tangshen Formula (TF) in rats with diabetic nephropathy. Methods All Wistar rats (n = 50) were randomly divided into the blank group, model group, monopril group (0.83 mg/kg), high-dose TF group (2.67 g/kg) and low-dose TF group ( 1.33 g/kg, each n = 10). The rat model of diabetic nephropathy was established by right-kidney nephrectomy and intraperitoneal injection of streptozotocin and then given the relevant drugs respectively for 20 weeks. The body weight, levels of blood glucose and ratio of urinary protein to creatinine were determined dynamically. When the trial was finished blood samples were taken from the abdominal aorta and plasma viscosity, the levels of total serum protein ( TSP), albumin, total cholesterol (TC), triglyceride (TG), blood urea nitrogen and creatinine were detected. The kidney tissue was taken for histopathological observation and lesion grade. Results Comparing with the model group, in the low-dose TF group the body weight increased significantly from 4th week ( P 〈 0.05 ), and much significantly from 12th week (P 〈0.01 ). In the high-dose TF group the ratio of urinary protein to creatinine decreased significantly ( P 〈 0.05 ), the levels of plasma viscosity ( P 〈 0.05 ), TC ( P 〈 0.05 ) and TG ( P 〈 0.01 ) decreased significantly, and glomerulosclerossi index (P 〈0. 01 ) and tubular fibrosis index (P 〈 0. 01 ) reduced. The results of the high-dose TF group were better than those of the monopril group. Conclusion Tangshen Formula has a good renal protective effect in rats with diabetic nephropathy. The mechanism may be related to that it can relieve the disorders of hemorrheology and blood lipid metabolism.
