目的 :应用17β雌二醇(E2)作用于大鼠骨髓间充质干细胞,研究其对干细胞体外诱导成脂向分化的影响。方法:分离、培养原代大鼠骨髓间充质干细胞(rat bone mesenchymal stem cells,r BMSCs),成脂诱导过程中加入10-9mol/L E2进行干预,采用油红O染色、实时荧光定量PCR、Western印迹等方法检测E2对干细胞脂向分化能力的影响。采用SPSS19.0软件包对数据进行统计学分析。结果 :10-9 mol/L E2能显著抑制大鼠BMSCs内脂滴形成,并使成脂相关基因(C/EBPα、C/EBPβ、PPARγ、a P2及ARDP)表达显著降低,PPARγ及ARDP蛋白表达下降。结论 :10-9 mol/L 17β雌二醇能显著抑制大鼠BMSCs成脂向分化。
PURPOSE: In this study, 10-9 mol/L 17 β-estradiol (E2) was applied in the adipogenic differentiation of rat bone mesenchymal stem cells (rBMSCs) and the effect of E2 was explored. METHODS: Rat BMSCs were obtained from the femurs and tibias of SD rats. 10-9 mol/L E2 was involved in the adipogenic differentiation of rBMSCs. Oil red staining, real time PCR and Western blot were carried out to detect the effect of 10-9 mol/L E2 on adipogenie differentiation of rBMSCs. The data was statistically analyzed using SPSS 19.0 software package. RESULTS: The use of 10-9 mol/L E2 decreased the amount of lipid droplets in rBMSCs and weakened the expression of adipogenie related genes and proteins like C/EBP α, C/EBP β, PPAR γ, aP2, and ARDP, which were significantly lower than the adipogenic induced group. CONCLUSIONS: The use of 10-9 mol/L E2inhibited adipogenie differentiation of rBMSCs significantly in vitro.
