新疆维吾尔族和汉族变应性鼻炎与CD14基因的相关研究

目的:研究新疆地区维吾尔族、汉族CD14基因rs2569192(C/G)和rs3138078(-1359G/T)二个位点多态性的分布特点及其与变应性鼻炎(AR)的关系。方法:选取新疆地区维吾尔族、汉族AR患者300例(AR组)与正常对照300例(对照组)。对600例样本进行CD14基因rs2569192和rs3138078位点SNP分型,并比较不同基因型的血清总IgE水平。结果:①新疆地区维吾尔族、汉族CD14基因rs2569192(C/G)和rs3138078(-1359G/T)二个位点基因型分布均以CC和GG型多见,以C和G等位基因为主。维吾尔族和汉族2个多态性位点分布差异无统计学意义(P〉0.05);②新疆地区维吾尔族、汉族AR组与对照组比较2个位点基因型和等位基因分布频率差异无统计学意义(P〉0.05)。维吾尔族AR组与汉族AR组比较rs2569192(C/G)位点基因型和等位基因分布频率差异有统计学意义(P〈0.05);③新疆地区维吾尔族、汉族人群rs2569192(C/G)位点多态性与北京汉族、日本、欧洲和非洲人群基因型和等位基因频率相比差异有统计学意义(P〈0.05);④新疆地区维吾尔族、汉族AR组与对照组比较,血清中总IgE明显偏高,差异有统计学意义(P〈0.05)。不同基因型与IgE水平无关。结论:①新疆地区维吾尔族、汉族都存在CD14基因rs2569192(C/G)和rs3138078(-1359G/T)多态性,都以CC、GG基因型和C、G等位基因为主,维吾尔族和汉族之间差异无统计学意义;rs2569192(C/G)位点分布有别于其他地区人群;②未发现CD14基因rs2569192(C/G)和rs3138078(-1359G/T)位点多态性与维吾尔族、汉族AR发病有关;③新疆维吾尔族、汉族AR患者血清总IgE明显高于正常人;CD14基因rs2569192和rs3138078位点不同基因型与IgE水平无关。

英文摘要:

Objective:This study aimed to investigate the relationship between CD14 gene rs2569192(C/G),rs3138078(-1359G/T) and allergic rhinitis(AR) in Xinjiang Uygur and Han populations as well as to determine characteristics of polymorphisms.Method:A total of 300 AR and 300 healthy controls subjects were included.The frequencies of genotypes and alleles were detected as well as the levels of tIgE in different genetypes were compared.Result:①The distribution of genotypes or alleles of CD14 gene rs2569192(C/G),rs3138078(-1359G/T) had no differences between the Xinjiang Uygurs and Hans(P0.05).The highest frequency of alleles was C,G.②The frequencies of genotypes and alleles were not different between the AR and control group in Uygur and Han(P0.05).The frequencies of genotypes and alleles of rs2569192 were different between the Uygur AR and Han AR group(P0.05).③The distribution of genotype frequencies and allele of rs 2569192 in the Xinjiang Uygur and Han population were quite different from Chinese Beijing Han populations,Japanese,European and African(P0.05).④The serum total IgE level in AR group was higher than that in healthy control group(P0.05).Conclusion:①rs2569192(C/G),rs3138078(-1359G/T) polymorphisms were not different between the Chinese Xinjiang Uygur and Han population.The major allele were both C and G.rs2569192 of CD14 in Xinjiang populations was different from that in the other populations.②No relationship between rs2569192,rs3138078 and AR was found.③The serum total IgE level in AR group was higher than that in healthy control group.No relationship between CD14 two SNP and serum total IgE level was found.
