目的研究氟对成纤维细胞Ca^2+浓度的影响并探讨其意义。方法应用扫描共聚焦显微技术测定不同染氟条件下(0.000l、l、10mg/LF^-)成纤维细胞Ca^2+水平的变化。结果染氟lmg/L组成纤维细胞在染氟500s左右时[Ca^2+]i水平开始升高,约l500s达到高峰,然后开始下降,3000s左右回到起点位置。结论成纤维细胞染氟lmg/LF^-组[Ca^2+]i有一过性升高。认为这种[Ca^2+]i浓度的变化在影响成纤维细胞中cbfal和某些生长因子之间的相百关系、但讲细朐增殖、分似两成骨嘉删转拯中可能起雷季作用。
Objective To study the effects of fluoride on the level of Ca^2+ release in cultured fibroblast. Methods Fibroblasts were cultured and loaded with fluo - 3 - AM. [ Ca^2+ ] i was measured by fluorescent intensity in each cell with laser confocal microscope. Results In the group of 1 mg/LF- , the intercellular free calcium concentration in fibroblasts increased transitorily. Conclusions The elevation of [ Ca^2+ ] i in fibroblast may play a important role in regulating the cooperation and restriction among the cbfal and other growth factors ,promoting the proliferation, differentiation, and the expression of osteogenesis phenotype.
