目的采用超高效液相色谱-四级杆-飞行时间串联质谱(UHPLC—Q—TOF/MS)法对红参醇提液中各成分进行分析和鉴定。方法色谱分离采用Poroshell120EC—C18色谱柱(3.0mm×100mm,2.7μm),以0.1%甲酸水溶液(A)-乙腈(B)为流动相梯度洗脱,流速为0.4mL·min-1;采用双电喷雾离子源(DualESI),在负离子模式下采集数据;一级质谱提供精确的分子量信息,误差限制在5ppm以内,得到该分子量最适的元素组成;二级质谱提供裂解碎片信息,与相关数据库和相关文献进行比对,推测其可能的化学结构。结果初步鉴定了红参醇提液中55个成分,包括原人参二醇型皂苷(PPD)27个,原人参三醇型皂苷(PPT)12个,齐墩果烷型皂苷1个,其他类成分15个。其中,10个化合物尚未有相关文献报道,结构需进一步确认。结论该研究丰富了对红参中化学物质的认识,为红参药效物质研究奠定了基础。
Objective To analyze and identify the chemical constituents of Radix Ginseng Destillata alcohol extract by ultra-high performance liquid chromatography-quadrapole-time of flight mass spectrometry (UHPLC-Q-TOF/MS). Methods The chromatographic separation was performed on an Agilent Poroshell 120 EC-Cls column (3.0 mm × 100mm, 2.7 μm) , 0.1% formic acid(A) and acetonitrile(B) were used as the mobile phase for gradient separation, and flow rate was set at 0.4 mL ·min-1. The constituents were detected in negative mode by Dual ESI-Q-TOF mass spectrometry. The MassHunter calculated the most suitable formula according to accurate molecular weight from MS, and the error can be limited within 5 ppm. The MS spectrometry provided fragmentation, and when the fragmentation was combined with reference data and related database, the possible structure of these constitutes were identified. Results A total of 55 compounds in Ginseng alcohol extract were primarily identified, including 27 protopanaxadiol type ginsenosides, 12 protopanaxatriol type ginsenosides, one oleanolic acid type ginsenoside and 15 other kinds of compounds via UHPLC-Q-TOF/MS. Conclusion To some extent, this study provides a foundation for the research of effective substance from Radix Ginseng Destillata.
